Amorphous Ground Substance (AGS) is a component of the extracellular matrix (ECM) in animal tissues. It is primarily composed of water, but also includes three major types of biomolecules: proteoglycans, glycosaminoglycans (GAGs), and multiadhesive glycoproteins.

  1. Proteoglycans: These are proteins that are heavily glycosylated. The basic proteoglycan unit consists of a core protein with one or more covalently attached glycosaminoglycan (GAG) chain(s).

  2. Glycosaminoglycans (GAGs): These are long unbranched polysaccharides containing a repeating disaccharide unit. The disaccharide units contain an amino sugar along with uronic sugar or galactose. GAGs are highly polar and attract water. They are thus useful to the body as a lubricant or shock absorber.

  3. Multiadhesive glycoproteins: These proteins contain multiple domains that allow them to bind to cells and other components of the ECM. They include fibronectin and laminin.

The AGS fills the space between cells and fibers in the ECM, helping to resist compressive forces on tissues. It also provides a medium for the exchange of nutrients and waste between cells and the bloodstream. The high water content and presence of GAGs give the AGS its characteristic gel-like consistency, which helps to provide lubrication in the joints and gives resilience to cartilage, for example.

The AGS also plays a role in cell signaling, with the multiadhesive glycoproteins helping to organize cells into tissues and coordinating cellular processes such as cell division and movement.
